Hh signaling is controlled by two receptors, Patched (Ptc) and Smoothened (Smo), on the target cell membrane. The Ptc can directly bind to the ligand and negatively regulate the Hh signal. The Smo is homologous to the G protein-coupled receptor, the N-terminus is located extracellularly, the C-terminus is located in the cell, the amino acid sequence of the transmembrane region is highly conserved.The serine and threonine residues at the C-terminus are phosphorylation sites, and the protein kinases catalyze the binding of phosphate groups. Members of this protein family have the function of transcription promoter only when maintaining the full length, and initiate transcription of the downstream target gene.when the carboxy terminus is hydrolyzed by the proteasome, a transcriptional repressor is formed to inhibit transcription of the downstream target gene. Smo is a receptor necessary for Hh signaling. In the absence of Hh, Ptc, activation of Smo can lead to activation of the Hh target gene. when the Smo is mutated, the same characterization as the Hh gene mutation can occur.
