PI3K-Akt Pathway
The serine/threonine kinase Akt acts as a proto-oncogene that plays an important role in the regulation of various cellular functions including metabolism, growth, proliferation, survival, transcription, and protein synthesis. Factors that activate the Akt signaling cascade, including receptor tyrosine kinases, integrins, B-cell and T-cell receptors, cytokine receptors, G-protein coupled receptors, and others capable of passing phosphatidylinositol III Kinase (PI3K) induces stimulation of triphosphate phosphatidylinositol (PIP3) production. Direct binding of Ras and p110 can activate PI3K.Activated PI3K produces PIP3.PIP3 binds to the PH domain-containing signaling protein Akt and PDK1, alters the protein structure of Akt, and translocates it to the cell membrane, allowing protein phosphorylation of PDK21 and PDK-2 on the membrane. Phosphorylation of PDK1 leads to Akt activation. Activated Akt activates or inhibits a series of downstream substrates such as Bad, caspase 9, NF-κB, GSK23 , thereby regulating cell proliferation, differentiation, apoptosis and migration.
